Recombinant Human VEGF
Source: A DNA sequence encoding the 165 amino acid
residue variant of human VEGF (Leung, D.W. et al., 1989, Science 246:1306) was expressed
in Sf 21 cells using a baculovirus expression system and the protein purified
by sequential chromatography.
Purity & Assay Method: >97%, as determined by SDS-PAGE,
visualized by silver stain
Formulation: Lyophilized from 62.1 µL of a 0.2
µm filtered solution in 30% acetonitrile, 0.1% TFA containing 50 µg of
bovine serum albumin per 1 µg of cytokine
Mass Per Vial: The vial size is 10 µg of recombinant human VEGF
Activity & Assay Method: The biological activity
of recombinant human VEGF was measured by its ability to stimulate 3H-thymidine
incomporation in human umbilical vein endothelial cells (Conn, G. et al.,
Proc. Natl. Acad. Sci. USA, 87:1323-1327). The ED50 for this effect
is typically 1-6 ng/mL.
Reconstitution: Reconstitute with sterile PBS containing
at least 0.1% HSA or BSA. The rhVEGF basic concentration in the stock solution should
be no less than 1 µg/mL.
Storage Conditions: Lyophilized samples are stable
for greater than six months at -20ºC to -70ºC. Upon reconstitution, this
cytokine can be stored under sterile conditions at 2º to 4ºC for one month
or at -20ºC to -70ºC for three months without detectable loss of activity.
AVOID REPEATED FREEZE-THAW CYCLES.
Sterilization: VEGF was filtered through a 0.2 micron
membrane and packaged under aseptic conditions.
Endotoxin Level & Assay Method: The endotoxin level
was determined by the LAL method and found to be less than 0.1ng per 1µg of
the cytokine.
