Strain Description: This Ptch1 conditional allele has been used to generate the pediatric malignant brain tumor Medulloblastoma (PMID 19213072) as well as the pediatric soft tissue sarcoma embryonal rhabdomyosarcoma (PMID 21316601). The allele is structural similar to the germline knockout allele of Ptch1 generated by Matthew P. Scott (PMID 9262482 ), which has been used in subsequent publications to generate medulloblastoma, rhabdomyosarcoma and basal cell carcinoma.

Cryopreservation Mating Scheme: The cryopreservation mating scheme is C57BL/6 males x heterozygous females. When maintained as a live colony, breeder pairs were distributed as a heterozygote female x wildtype C57BL/6 male. According to information received from the donating investigator, Dr. Charles Keller, experimental animals are cancer-prone in the heterozygous (haploinsufficient) state. Heterozygous males may be used to expand the colony; however, for experimentals, the Ptch1 conditional allele ALWAYS comes from the dam since Cre comes from the sire. The strain being distributed is the conditional Ptch1, not cre. Average litter size is 5-8.
