Strain Description: Male and female Aprt(+/neo) or Aprtneo/neo mutant mice of 129/Sv background are crossed to wild-type mice of another strain to generate Aprt(+/neo) F1 hybrids. With time, LOH occurs at the Aprt locus in some cells. These cells can be isolated through selection for the loss of Aprt. By analyzing Aprt gene sequence, simple sequence repeat markers flanking the Aprt locus on chromosome 8, and cytological markers, it is possible to assign each somatic cell variant to a particular LOH pathway. The Aprt heterozygous model has been extremely useful in dissection of mutational pathways in vivo and in stem cells. Among the findings is that loss of heterozygosity (LOH) is the predominant pathway to mutation in vivo but is suppressed by about two orders of magnitude in stem cells.
