Antitumor effects of nonconjugated murine Lym-2 and human-mouse chimeric CLL-1 monoclonal antibodies against various human lymphomas

    Lym-2 is a murine monoclonal antibody(moAb) reactive with both normal and neoplastic human lymphocytes directed towards a human class II molecule variant. Previous studies have demonstrated that signals transmitted by class II molecules which stimulate normal lymphocytes can be inhibitory for B-cell lymphoma growth by inducing activation-induced cell death. We evaluated the effects of nonconjugated murine Lym-2 on the growth of various human lymphomas using both in vitro and in vivo assays. A human-mouse chimeric Lym-2 (ch-CLL1) was also examined. Cell lines derived from Burkitt's lymphoma, diffuse large cell B-cell lymphoma, anaplastic large cell lymphoma, and EBV-induced B-cell lymphomas were incubated with Lym-2. Lym-2 was capable of directly inhibiting the growth of a variety of human lymphoma cell lines in vitro and this inhibitory effect could be further enhanced by prior crosslinking of the antibody. These human lymphomas were then transferred into mice with severe combined immunodeficiency (SCID). Treatment with either murine or a human-mouse chimeric Lym-2 were equally effective in significantly improving the survival of tumor-bearing mice. These results indicate that stimulation by nonconjugated human-mouse chimeric Lym-2 may offer a biologic approach to the treatment of human lymphomas.
