Laboratory Overview

The Laboratory of Molecular Technology is an integrated, high-throughput molecular biology laboratory focusing on the development of genetics and genomics technologies, together with associated laboratory automation systems, data analysis, and information management tools. The laboratory develops integrated strategies using multiple technology platforms that maximize the value of clinical samples and lead to the identification of pathways, genes, or gene products involved in the development of disease, as well as the assembly of genetic or expression profiles for the identification of prognostic and diagnostic indicators. The laboratory also specializes in the development of custom assays for quantitation of specific molecules in complex clinical samples. Sequencing and Automation

The LMT provides DNA sequencing on several different platforms.  Sanger sequencing capabilities range from one to several thousand sequences.  Next generation sequencing (NGS) is available on the Roche/454 platform and the Ion Torrent PGM.  The LMT has a variety of options for NGS library preparations and provides substantial project design consultation. The LMT also provides specific single nucleotide polymorphism (SNP) detection and DNA methylation analysis on the Qiagen Pyromark platform.  Large projects requiring laboratory automation are managed using one of several Beckman BioMek FX liquid handling systems.

Molecular Diagnostics

The LMT provides Sanger sequencing-based mutation detection services in a CLIA environment for investigators evaluating patients for specific genetic abnormalities. We have established a large panel of mutation detection assays and will  develop new assays on request.  Current assays can deliver results in 1-3 weeks and new assays can be developed within 6 weeks.

Microarray  and Expression Services

A large variety of services to measure gene expression and DNA copy number variations are available from the LMT.  We deliver Affymetrix services that include single arrays and high throughput formats. We provide array-based comparative genomic hybridization on Agilent microarray platforms.  Measurement of expression for focused gene sets and miRNA can be carried out on the Nanostring platform.

Genotyping and Quantitative PCR

A number of  qPCR and genotyping assays on several different platforms are available.  The LMT can give guidance on the best platform and chemistry as well as manage all project aspects from assay design to data analysis. The laboratory has considerable experience with many assays using ABI’s TaqMan chemistry from copy number variation assays to SNP assays to quantitative PCR assays.  The Fluidigm microfluidics platform is a great choice for projects requiring  large numbers of gene expression measurements.  The LMT also offers customized assay design for all types of qPCR assays.
