Surface capture of tagged proteins
The PCL is collaborating with PEL to develop a strategy to identify the optimal affinity tag with which to capture a target protein on to a sensor chip surface. Our expectation is that this will be dependent on the protein of interest. We have expressed two proteins of interest with the following peptide and protein tags; Strep2, His 6, His 9, His 12, GST, MBP and ZZ. The effect on expression levels will be reported, capture affinity and also binding affinity of a ligand to the protein of interest.
