Improving protein expression in the yeast, Kluyveromyces lactis

The main goal of this technology development effort in the Protein Expression Laboratory (PEL) is to evaluate protease deficient strains of Kluyveromyces lactis for improving protein expression.  One of the main problems with protein expression from many hosts is protein degradation from host proteases.  This effort determined that some strains improve protein quality compared to others and for that reason we routinely test multiple strains for protein quality for each new protein expression project.

Six differProteolysis in different K. lactis strainsent protease knockout strains were tested for reduced degradation of the same protein after 48 total hours of growth (24 hours of growth in induction medium).
