Lymphokine Testing Section

The laboratory offers testing of cytokines and other soluble factors using standard sandwich ELISA assays, multiplex assays based on Meso Scale Discovery kit assays and cytokine bioassays. The recent addition of the Meso Scale Sector 6000 imager also provides the capacity to perform tests for a wide variety of other markers in single or multiplex formats. These include tests for phosphoproteins, vascular factors, cardiac and metabolic factors. Contact laboratory staff for further information.

Multiplex Assays

The laboratory recently was able to acquire a Meso Scale Discovery Sector 6000 instrument for multiplexing a variety of assays, including cytokines. The instrument uses standard plate formats containing 1, 4, 7 or 10 products in a single 96 well assay and 1 or 4 products in a 384 well plate format. Sample volume requirements are significantly lower than ELISA assays (25ml per well in a 96 well format) and the instrument is compatible with high throughput testing with read times for 96 well plate assays of under 3 minutes. Kits are available for cytokine testing as well as for other markers. Reagents are also available for use in assay development or for production of custom single plex assays. It is recommended that all samples be microfuged prior to testing to remove particulates. The Meso Scale platform uses an electrochemiluminescence detection system that is designed to minimize nonspecific reactions and background noise. All pricing is based on full plate assays testing 37 samples per plate in duplicate wells in a 96 well plate format and M&S costs are listed separately from labor. Custom plates can be requested but require a commitment to purchase a minimum of 10 plates.

ELISA Assays

ELISA kit based assays are normally provided in a format of 12 strips of 8 wells (96-well). Costs are based on the number of strips used for testing and for the assays listed below kits are obtained from commercial sources. Testing is performed in duplicate wells and two strips are required per kit for generation of a standard curve. The kit M&S costs are divided into multiple tiers based on kit price. Additional materials include controls for monitoring kit performance and for quality control. Labor costs are also added based on half plate or full plate assays with some labor discount associated with multiplate assays. Total cost for these tests is the sum of the cost of test strips required + fixed materials costs + labor. Due to the cost of standards and controls, the cost per sample is lowest when full plate testing is performed.

Cytokine Bioassays

The LTS has expertise in a number of bioassay systems for measuring cytokine levels. These include assays for IL-2, IL-4, IL-6, IL-7, IL-3/GMCSF/GCSF, human and mouse interferon and TNF. Please contact the laboratory staff to discuss specific requirements. While general pricing has been developed for these assays, the scope of individual requests vary significantly.
