Influenza virus-assisted entry of a TCL-tropic HIV-1 isolate into CD4+ cells

    We investigated the ability of human immunodeficiency virus type 1 (HIV-1) to infect CD4+ cells which were previously infected with an influenza virus strain (A/PR/8H1N1). Sub-confluent monolayers of cells were infected with influenza virus at 37 degrees C and subsequently tested for their susceptibility to HIV-1 infection by the detection of early and late transcripts in the virus life cycle at 0, 2 and 24 hours at 37 degrees C. CD4+ non-human cells (mink-CD4) became susceptible to HIV-1 entry when they were previously infected with influenza virus. Influenza virus-infected human CD4+ cells (primary human CD4+ and HeLa-CD4) showed significant enhancement of HIV-1 entry and replication. Influenza virus-mediated HIV-1 entry was only observed for a T-cell tropic HIV-1 (NL4-3). Entry of a macrophage-tropic HIV-1 strain (US1) was not influenced by prior infection of the target cells with influenza virus. This selectivity might be mediated by a significant increase in cell surface expression of TCL-tropic coreceptor CXCR4 in HeLa-CD4 cells, as shown by immunofluorescence. Our observations may be significant in understanding the mechanisms of a shift from M-tropic to TCL-tropic HIV-1 phenotype associated with occurrence of opportunistic infections in HIV-1 infected individuals.
